[目的]研究选择性环氧合酶-2(COX-2)抑制剂尼美舒利对人结肠腺癌细胞Colo-320增殖凋亡的影响及机制。[方法]MTT法检测经不同浓度处理的Colo-320细胞增殖率,流式细胞术检测其凋亡率,RT-PCR及Western Blot法检测COX-2、Bcl-2、VEGF的mRNA、蛋白表达率。[结果]经50、100、200、300μmol/L尼美舒利处理的Colo-320细胞增殖率分别为0.98±0.16,0.95±0.09,0.51±0.06和0.48±0.08,凋亡率分别为9.52±0.41、12.11±0.71、28.13±3.65和39.37±4.15。药物处理后的Colo-320细胞中COX-2的mRNA、蛋白表达在各组间无统计学差异(P>0.05),Bcl-2及VEGF mRNA、蛋白表达在高浓度组较低浓度组、对照组明显降低(P<0.01)。[结论]尼美舒利可能通过下调抗凋亡因子(Bcl-2)及血管内皮生成因子(VEGF)表达来抑制Colo-320细胞增殖、诱导凋亡。
[Objective] To investigate the effect and mechanism of nimesulide, a selective cyclooxygenase-2 (COX-2) inhibitor, on proliferation and apoptosis of human colon adenocarcinoma cell line Colo-320. [Methods] The proliferation rate of Colo-320 cells treated with different concentrations was detected by MTT assay and the apoptosis rate was detected by flow cytometry. The mRNA and protein expression of COX-2, Bcl-2 and VEGF were detected by RT-PCR and Western Blot rate. [Result] The proliferation rates of Colo-320 cells treated with 50, 100, 200 and 300μmol / L nimesulide were 0.98 ± 0.16, 0.95 ± 0.09, 0.51 ± 0.06 and 0.48 ± 0.08 respectively, and the apoptosis rates were 9.52 ± 0.41, 12.11 ± 0.71, 28.13 ± 3.65 and 39.37 ± 4.15. The mRNA and protein expressions of COX-2 in Colo-320 cells after treatment were not significantly different among the three groups (P> 0.05). The mRNA and protein expressions of Bcl-2 and VEGF were lower in high concentration group than those in low concentration group Group was significantly lower (P <0.01). [Conclusion] Nimesulide may inhibit the proliferation and induce apoptosis of Colo-320 cells by down-regulating the expression of Bcl-2 and VEGF.
