目的:探讨滋补肝肾复方复健片对局灶性脑梗死大鼠颈髓Rho相关卷曲螺旋形成蛋白激酶2(ROCK2)的作用。方法:采用右侧近端大脑中动脉电凝术制备大脑中动脉阻塞(MCAO)模型。动物随机分为假手术组、模型组、药物组,其中3组又各分为术后4周组和术后5周组,每组各8只。药物组按9g生药/kg给予复健片水溶液,其余两组分别给予同等量0.9%氯化钠溶液,1次/d。分别采用RT-PCR和Western blot方法检测大鼠左侧颈髓ROCK2 mRNA和蛋白的表达。结果:术后4周末,模型组与药物组的ROCK2 mRNA和蛋白表达均较假手术组显著性降低(P<0.05,P<0.01),而药物组较模型组降低更为显著(P<0.01),至术后5周末;模型组与假手术组ROCK2 mRNA和蛋白表达已无显著差异,但药物组仍然显著低于其余两组(P<0.01,P<0.05)。结论:复健片可抑制MCAO大鼠失神经支配侧颈髓ROCK2基因和蛋白的表达,从而有助于促进局灶性脑梗死皮质脊髓束重塑。
Objective: To investigate the effect of Nourishing Liver and Kidney Complex Tablets on Rho-associated coiled-coil protein kinase 2 (ROCK2) in the spinal cord of rats with focal cerebral infarction. Methods: The middle cerebral artery occlusion (MCAO) model was established by right proximal middle cerebral artery electrocoagulation. The animals were randomly divided into sham-operation group, model group and drug group, of which 3 groups were divided into 4 weeks after operation and 5 weeks after operation, 8 rats in each group. Drug group by 9g crude drug / kg rehabilitation tablets aqueous solution, the other two groups were given the same amount of 0.9% sodium chloride solution, 1 time / d. The expression of ROCK2 mRNA and protein in the left cervical spinal cord were detected by RT-PCR and Western blot respectively. Results: After 4 weeks of operation, the expression of ROCK2 mRNA and protein in the model group and the drug group were significantly lower than those in the sham operation group (P <0.05, P <0.01), while those in the drug group were significantly lower than those in the model group (P <0.01) ) To 5 weeks after operation. There was no significant difference in ROCK2 mRNA and protein expression between the model group and the sham operation group, but the drug group was still significantly lower than the other two groups (P <0.01, P <0.05). CONCLUSION: Fufang Tablet can inhibit the expression of ROCK2 gene and protein in the denervated innervated cord of MCAO rats, which may be helpful to promote the remodeling of corticospinal tract in focal cerebral infarction.
