目的 制备HLA -B2 7单克隆抗体。方法 用HLA -B2 7抗原免疫小鼠 ,取脾细胞与骨髓瘤细胞融合制备杂交瘤细胞 ,用ELISA法筛选分泌该单抗的细胞株。结果 建立了分泌该单抗的细胞系 3B3和 4F2 ,抗体均为IgG2b ,3B3细胞分泌的单抗只与B2 7反应 ,而 4F2细胞分泌的单抗与B7有交叉反应。 结论 建立了高特异性分泌抗B2 7单抗的细胞株 ,具有潜在的应用价值
Purpose To prepare HLA-B2 7 monoclonal antibody. Methods Mice were immunized with HLA-B2 7 antigen. The spleen cells and myeloma cells were fused to prepare hybridoma cells. The cell lines secreting the McAb were screened by ELISA. Results The McAbs 3B3 and 4F2 secreting the McAb were established. The antibodies were all IgG2b. The mAbs secreted by 3B3 cells only reacted with B2 7, while the McAbs secreted by 4F2 cells cross-reacted with B7. Conclusion A highly specific cell line secreting anti-B2 7 monoclonal antibody was established, which has potential application value
