三七总皂苷动员心肌梗死大鼠骨髓干细胞出髓效率的实验研究

目的:观察三七总皂苷对急性心肌梗死(acute myocardial infarction,AMI)大鼠骨髓干细胞(bone marrow stemcells,BMSC)动员的效率。方法:140只Wistar大鼠建立急性AMI模型后随机分成:西药动员组(皮下注射G-CSF50μg.kg-1.d-1;假手术组和模型组皮下注射生理盐水50μg.kg-1.d-1;中药动员组腹腔注射血塞通150 mg.kg-1.d-1;中西药动员组皮下注射G-CSF50μg.kg-1.d-1和腹腔注射血塞通150 mg.kg-1.d-1,除假手术组外,各组又按1,7,14 d 3个时间点分为3个亚组,其中G-CSF每天1次,共用7 d,血塞通注射液每天1次,直到每亚组处死动物前停止,采用流式细胞仪检测外周血中C-kit+细胞量和全自动血细胞分离机观察白细胞(white blood cell,WBC)计数和MNC(mononuclear cell,MNC)百分比。结果:模型组与假手术组1,7,14 d比较WBC、MNC%明显升高(P<0.05);在1,7,14 d中药组、西医药组与同期中西医药组相比明显低(P<0.05);西药组、中西医药组与模型组相比外周血单个核细胞C-kit受体表达明显升高(P<0.05),中西药组与各组相比变化明显。结论:三七总皂苷可协同G-CSF更好动员BMSC,提高BMSC出髓效率,增加外周血中BMSC数量。 OBJECTIVE: To observe the efficiency of Panax notoginseng saponins in the mobilization of bone marrow stem cells (BMSCs) in rats with acute myocardial infarction (AMI).
