目的:观察四六汤对黄褐斑模型的影响,探讨四六汤防治黄褐斑的药效学作用。方法:选取昆明种雌性小鼠60只,将动物按数字随机分成6组。分别为空白对照组、模型对照组、四六汤0.36g/kg,0.72 g/kg,1.44 g/kg剂量组、维生素C 0.1g/kg。采用中波紫外线照射雌性小鼠复制黄褐斑实验动物模型,取其受试脱毛后皮肤组织标本,严格按酪氨酸、MDA(丙二醛)、SOD(超氧化歧化酶)试剂盒说明书进行指标测定。另外取小鼠去毛皮肤1块(1.0×1.0cm),10%福尔马林固定,常规石蜡包埋切片,用HMB45(melanoma黑素瘤)为第一抗体,免疫组化二步法进行病理形态学的检测。结果:模型对照组小鼠与空白对照组相比,皮肤中酪氨酸含量和MDA明显升高;SOD活性明显降低,血清中雌二醇(E2)、孕酮(P)含量明显升高,睾酮(T)含量明显降低;靶目标内黑色素阳性细胞的个数、平均光密度和数密度均明显增多;黑色素阳性细胞平均灰度明显下降。四六汤1.44 g/kg剂量组与模型组比较,酪氨酸含量、MDA、P、E2明显降低,SOD活性、T含量明显升高,皮肤中黑色素细胞个数、数密度和平均光密度明显下降,黑色素阳性细胞平均灰度明显升高。结论:四六汤防治黄褐斑的作用可能是与抗氧化,减少皮肤组织中酪氨酸、MDA含量,提高SOD活性,有效抑制皮肤黑素细胞增殖,从而减少黑素合成有关。

Compared with the model group, the content of tyrosine, MDA, P and E2 of Siliari Decoction 1.44 g / kg dose group decreased obviously, SOD activity and T content increased obviously, and the number of melanocytes, number density and average optical density in skin were significantly Decreased, the average gray level of melanin-positive cells increased significantly. Conclusion: The effect of Siwu Decoction on the prevention and treatment of melasma may be related to anti-oxidation, reducing the content of tyrosine and MDA in skin tissue, increasing the activity of SOD and inhibiting the proliferation of melanocytes in the skin, thereby reducing the melanin synthesis.
