为调查酒精性肝硬化(AC)体内外单核细胞肿瘤坏死因子(TNFa)和可溶性肿瘤坏死因子受体(sTNFR)的表达,我们采用酶联免疫方法对32例AC血清及12/32例周围血单核细胞自发性和经乙醇、内毒素酯多糖(LPS)刺激后TNFα和sTNFR的表达进行了检测。结果与结论:血清中TNFα、sTNFRp55、sTNFRp75水平较正常对照组明显升高。与健康者比较,经LPS刺激后的单核细胞在AC组不论TNFα还是sTNFRp55、sTNFRp75都是升高的,而经乙醇刺激后TNFα和sTNRp55在两组之间没有差异。尽管sTNFRp75表达增高,但这种增高的表达在自然表达中就已经显示出来。这些结果提示AC单核细胞活化与TNFα和sTNFR的表达与内毒素有关。TNFα和sTNFR的检测对AC发生机理,病情发展和预后判断有重要意义。
To investigate the expression of monocyte-derived tumor necrosis factor (TNF) and soluble tumor necrosis factor receptor (sTNFR) in alcoholic cirrhosis (AC) in vitro and in vivo, we used enzyme-linked immunosorbent assay to detect 32 cases of AC serum and 12/32 cases The monocyte spontaneity and the expression of TNFα and sTNFR after stimulation with ethanol and endotoxin ester polysaccharide (LPS) were detected. RESULTS AND CONCLUSION: The levels of TNFα, sTNFRp55 and sTNFRp75 in serum were significantly higher than those in normal controls. Compared with healthy controls, mononuclear cells stimulated by LPS increased in sTNFRp75 and sTNFRp55 regardless of TNFα or sTNFRp55, while TNFα and sTNRp55 did not differ between the two groups after stimulation with ethanol. Despite the increased sTNFRp75 expression, this increased expression has been demonstrated in natural expression. These results suggest that activation of AC monocytes and expression of TNFα and sTNFR are associated with endotoxin. The detection of TNFα and sTNFR is of great significance to the pathogenesis and prognosis of AC.
