目的 探讨解偶联蛋白 3( U CP3)基因二核苷酸 ( CA) n串联重复序列多态标记与中国汉族人 2型糖尿病的关系。方法 用 PCR-变性聚丙烯酰胺凝胶电泳及银染方法 ,检测 2 0 8例南京地区汉族人 ( 2型糖尿病 10 6例 ,正常对照 10 2例 )U CP3基因的 ( CA) n串联重复序列多态标记 ,比较各组间的等位基因频率和基因型频率。结果 2型糖尿病组和正常对照组U CP3基因等位基因频率的分布存在显著性差异 ( χ2 =2 6 .12 ,P<0 .0 0 5 ) ;2型糖尿病组 UCP36、7、8等位基因频率显著高于正常对照组 ( RR=4.49,P<0 .0 0 5 ;RR=7.14,P<0 .0 0 5 ;RR=14.2 3,P<0 .0 5 ) ;中国南京地区汉族人群与高加索人群 UCP3基因等位基因频率的分布亦有显著性差异 (χ2 =37.43,P<0 .0 0 5 )。结论 U CP3基因 ( CA) n串联重复序列多态标记与中国汉族人 2型糖尿病相关联
Objective To investigate the relationship between the polymorphism of dinucleotide (CA) n tandem repeat (UCP3) gene and type 2 diabetes mellitus in Chinese Han population. Methods PCR-denaturing polyacrylamide gel electrophoresis and silver staining were used to detect the (CA) n tandem repeat of U CP3 gene in 208 Han Chinese patients (106 cases of type 2 diabetes mellitus and 102 cases of normal controls) Polymorphic markers were used to compare allele frequencies and genotype frequencies between groups. Results There was a significant difference in the distribution of allele frequencies of UCP3 gene in type 2 diabetes mellitus and controls (χ2 = 2.12, P <0.05).
