采用BALB/cnu+裸鼠 ,建立脐血移植的裸鼠模型 ,探讨分离、冻存后的脐血造血细胞在小鼠体内的造血活性及进入体内后的迁移、定居规律 ,为临床移植及大规模建立脐血库作准备。结果显示羟乙基淀粉沉降法分离及以DMSO为保护剂的阶段降温法冻存脐血造血细胞回收率高 ,活力好 ,每只小鼠输入 (1.0 - 2 .0 )× 10 7个脐血单个核细胞 ,就能在其体内查到成活的证据。结果表明 :①羟乙基淀粉沉降法和以DMSO为保护剂的阶段降温法能有效地回收脐血造血细胞和保持其造血活性 ,在临床移植和建立脐血库时可以采用。②人类脐血富含造血干细胞 ,能跨越种属障碍 ,在裸鼠体内移植成活。③脐血造血细胞进入裸鼠体内后 ,先散布于骨髓、肝、脾、肺、肾等脏器间 ,最后定居于骨髓。冻存可能影响造血细胞表面粘附分子 ,影响其定居行为 ,但并不影响其造血活性。

Human umbilical cord blood is rich in hematopoietic stem cells that can cross the species barrier and survive in vivo in nude mice. ③ cord blood hematopoietic cells into nude mice, first spread in the bone marrow, liver, spleen, lung, kidney and other organs, and finally settled in the bone marrow. Cryopreservation may affect the adhesion of molecules on the surface of hematopoietic cells, affecting their colonization behavior, but does not affect its hematopoietic activity.
