目的探讨植物花色苷矢车菊素-3-葡萄糖苷(Cy-3-g)对高胆固醇血症小鼠血小板颗粒物释放的影响。方法将7周龄雄性C57小鼠随机分成正常对照组、高胆固醇血症模型组和Cy-3-g干预组。正常对照组饲喂AIN-93G标准饲料,高胆固醇模型组饲喂高脂高胆固醇饲料,Cy-3-g干预组饲喂添加Cy-3-g(1 000 mg/kg)的高脂高胆固醇饲料。饲喂12周后,取小鼠血液测定血清总胆固醇(TC)的含量。确认模型建立成功后,心脏取血,分离纯化血小板悬液,测定各组小鼠血小板的三磷酸腺苷(ATP)释放、五羟色胺(5-HT)释放以及P选择素(CD62P)和CD40L在血小板表面的表达量。结果实验期末,高胆固醇血症组和Cy-3-g干预组的血清总胆固醇明显高于正常对照组。与高胆固醇血症组相比,干预组血小板的ATP释放、5-HT释放及CD62P和CD40L表达水平明显降低。结论花色苷Cy-3-g能够抑制高胆固醇血症小鼠血小板颗粒物释放,提示植物花色苷对心血管疾病具有一定的保护作用。
Objective To investigate the effect of plant cyanidin Cy-3-g on platelet particle release in hypercholesterolemic mice. Methods Seven-week-old male C57 mice were randomly divided into normal control group, hypercholesterolemia model group and Cy-3-g intervention group. The normal control group was fed with AIN-93G standard diet, the high cholesterol model group was fed with high fat and cholesterol diet, and the Cy-3-g intervention group was fed with high fat and high cholesterol with Cy-3-g (1000 mg / kg) feed. After 12 weeks of feeding, the blood of mice was taken to determine the content of serum total cholesterol (TC). After confirming the successful establishment of the model, the blood was taken from the heart and the platelet suspension was isolated and purified. The platelet ATP release, serotonin (5-HT) release and the expression of CD selectin (CD62P) and CD40L on the platelet surface the amount. Results At the end of the experiment, serum total cholesterol in hypercholesterolemia group and Cy-3-g intervention group was significantly higher than that in normal control group. Compared with hypercholesterolemia group, ATP release, 5-HT release and CD62P and CD40L expression levels of platelet in intervention group were significantly decreased. Conclusion Anthocyanin Cy-3-g can inhibit the release of platelet particles in hypercholesterolemia mice, suggesting that plant anthocyanins may have a protective effect on cardiovascular diseases.
