Krüppel样因子5(Krüppel-like factor 5,KLF5)是一类进化保守的锌指型转录因子,它能调控下游众多靶基因的表达。KLF5参与细胞增殖、分化和凋亡等细胞生物学过程。翻译后修饰对KLF5的稳定性和发挥作用产生影响,而KLF5与非编码RNA之间也存在相互作用。越来越多的研究表明,KLF5异常表达于人类多种肿瘤(包括结直肠癌、乳腺癌、前列腺癌等)。KLF5不仅参与肿瘤的发生,而且与患者预后相关。有趣的是,KLF5在一部分肿瘤中的功能表现为环境依赖性,而且翻译后修饰和性激素水平影响其功能的发挥。鉴于KLF5在肿瘤中的重要作用,它被认为是一个潜在的肿瘤诊断标志物和肿瘤治疗新靶点。
Krüppel-like factor 5 (KLF5) is a class of evolutionarily conserved zinc finger transcription factors that regulate the expression of many downstream target genes. KLF5 is involved in cell biology processes such as cell proliferation, differentiation and apoptosis. Post-translational modifications have an impact on the stability and function of KLF5, but there is also an interaction between KLF5 and non-coding RNA. More and more studies have shown that KLF5 is abnormally expressed in many human tumors (including colorectal cancer, breast cancer, prostate cancer, etc.). KLF5 not only involved in the occurrence of tumors, but also with the prognosis of patients. Interestingly, the function of KLF5 in some tumors is environmental-dependent, and post-translational modifications and sex hormone levels affect their function. In view of the important role of KLF5 in tumors, it is considered as a potential tumor diagnostic marker and a new target for tumor therapy.
