目的 探讨放射性核素90 Sr对成纤维细胞超微结构的影响 ,寻找出应用90 Sr防治瘢痕的最有效剂量。方法 用90 Sr敷贴器照射体外培养的成纤维细胞KMB— 17,照射量为 2 0 0— 10 0 0cGy ,然后获取实验组和对照组标本 ,再用JEM— 10 0CX透射电镜观察成纤维细胞的超微结构变化。结果 90 Sr2 0 0cGy、 6 0 0cGy照射KMB— 17,二组细胞都表现细胞体积大、核大、核仁明显、常梁色质丰富、胞质丰富 ,粗面内质网丰富、扩张 ,与对照组相比 ,6 0 0cGy组细胞功能更为活跃 ,2 0 0cGy组无明显差异 ,10 0 0cGy组细胞体积大 ,但胞浆内见大量游离核糖体 ,粗面内质网较少 ,扩张不明显 ,线粒体肿胀。结论 小、中剂量90 Sr照射可刺激成纤维细胞功能活跃 ,大剂量90Sr照射则抑制成纤维细胞功能
Objective To investigate the effect of radionuclide 90 Sr on the ultrastructure of fibroblasts and to find out the most effective dose of 90 Sr for preventing and treating scar. Methods 90 Sr applicator was used to irradiate KMB-17 fibroblasts cultured in vitro, and the irradiation dose was 200-1000 cGy. The experimental group and the control group were harvested, and the fibroblasts were observed with JEM-10OC transmission electron microscope Ultrastructural changes. Results 90 Sr2 0 0cGy and 6 0 0 cGy irradiated KMB-17 cells in both groups showed large cell volume, large nucleus, obvious nucleoli, abundant chromatin, abundant cytoplasm, rich and expanded rough endoplasmic reticulum, and Compared with the control group, the cell function in the 600 cGy group was more active, and there was no significant difference in the 200 cGy group. The cells in the 100 cGy group were bulky, but there were a large number of free ribosomes in the cytoplasm, with less rough endoplasmic reticulum and dilatation Obviously, mitochondria are swollen. Conclusion Small and medium doses of 90 Sr irradiation can stimulate fibroblasts active, high-dose 90Sr irradiation inhibit fibroblast function
