目的研究尾加压素Ⅱ(UⅡ)及G蛋白偶联受体14(GPR14)mRNA在人嗜铬细胞瘤组织中的表达。方法提取正常肾上腺皮质、髓质和嗜铬细胞瘤组织总RNA,采用逆转录-聚合酶链反应的方法,以扩增的目的条带灰度和内参照条带灰度比值作为检测指标,对UⅡ和GPR14mRNA的表达进行半定量分析。结果正常肾上腺皮质和髓质中UⅡ及GPR14mRNA表达差异无显著性;嗜铬细胞瘤组织中UⅡ及GPR14mRNA的表达显著低于正常肾上腺皮质和髓质(P<0.05);肾上腺嗜铬细胞瘤组织中GPR14mRNA的表达显著低于肾上腺外嗜铬细胞瘤组织(P<0.05)。结论UⅡ及GPR14mRNA在正常肾上腺和嗜铬细胞瘤组织中表达水平不同,提示其在嗜铬细胞瘤的发病机制和血压调节中可能起一定的作用。
Objective To investigate the expression of urotensin Ⅱ (UⅡ) and G protein-coupled receptor 14 (GPR14) mRNA in human pheochromocytoma. Methods The total RNA was extracted from normal adrenal cortex, medulla and pheochromocytoma by reverse transcription polymerase chain reaction (RT-PCR), and the gray value of target band and the gray value of internal reference band were taken as detection indexes. UII and GPR14 mRNA expression for semi-quantitative analysis. Results The expression of UⅡ and GPR14 mRNA in adrenal cortex and medulla had no significant difference. The expression of UⅡ and GPR14 mRNA in pheochromocytoma was significantly lower than that in normal adrenal cortex and medulla (P <0.05). The expression of UⅡ and GPR14 mRNA in pheochromocytoma GPR14 mRNA expression was significantly lower than that of adrenal pheochromocytoma (P <0.05). Conclusions The expression levels of UⅡ and GPR14 mRNA in normal adrenal gland and pheochromocytoma tissues are different, suggesting that they may play a role in the pathogenesis and regulation of pheochromocytoma.
