目的研究北沙参内生真菌的抗菌活性,鉴定活性真菌的分类地位。方法采用植物组织分离法从北沙参基原植物珊瑚菜Glehnia littoralis中分离内生真菌。以大肠杆菌、金黄色葡萄球菌和白色假丝酵母为指示菌株,采用牛津杯法研究北沙参内生真菌的抑菌活性,对具有抑菌活性的内生真菌进行分子鉴定。结果有4株内生真菌对大肠杆菌具有抑制作用,内生真菌发酵浓缩液抑菌圈直径与4 U/mL硫酸庆大霉素抑菌圈直径的比值(d/D)最大可达1.07;有15株内生真菌对金黄色葡萄球菌具有抑制作用,d/D值最大为0.65;有3株内生真菌对白色假丝酵母具有抑制作用,内生真菌发酵浓缩液与0.2 mg/mL氟康唑的d/D值最大可达1.27。19株具有抑菌活性的内生真菌归于4个目,5个科,7个属,8个种。结论北沙参植物中存在多种抑菌活性的内生真菌,对于北沙参内生真菌资源的开发及药材道地性的评价具有一定的指导意义。
Objective To study the antibacterial activity of endophytic fungi of Radix isatidis and to identify its taxonomic status. Methods Endophytic fungi were isolated from Glehnia littoralis, a primitive plant belonging to the genus Escherichia coli, Staphylococcus aureus and Candida albicans were used as indicator strains. The antibacterial activities of endophytic fungi of Radix iscaenidae were studied by the method of Oxford cup, and the endophytic fungi with antibacterial activities were identified. Results Four endophytic fungi had an inhibitory effect on Escherichia coli. The ratio of the inhibition zone diameter of endophytic fungi fermentation concentrate to the diameter of inhibition zone of 4 U / mL gentamycin sulfate (d / D) was up to 1.07. 15 strains of endophytic fungi have inhibitory effect on Staphylococcus aureus, and the maximum value of d / D is 0.65. Three endophytic fungi have inhibitory effects on Candida albicans. The endophytic fungus fermentation concentrate and 0.2 mg / mL fluoride The d / D value of the azole is up to 1.27.19. The endophytic fungi with antibacterial activity belong to 4 orders, 5 families, 7 genera and 8 species. Conclusion There are many antibacterial activity endophytic fungi in the plants of Radix isatidis, which is of guiding significance for the development of endophytic fungi resources and the evaluation of medicinal authenticity.
