为了研究锌7-金属硫蛋白(Zn7-metallothionein, Zn7-MT)对细胞内外过氧化氢引起的NIH3T3细胞增殖的影响, 本文采用了MTT细胞计数法,3H-TdR掺入实验,细胞周期时相测定等方法。结果显示低浓度的甲萘醌和过氧化氢促进了NIH3T3细胞增殖, 使其MTT计数增加,细胞周期S期比例增高,DNA合成率增加。锌7-金属硫蛋白本身对NIH3T3细胞增殖没有影响,但它可以抑制甲萘醌和过氧化氢对NIH3T3细胞促增殖作用。提示锌7-金属硫蛋白对预防由活性氧诱导的成纤维细胞增殖所引起的疾病可能具有重要作用。
In order to study the effect of Zn7-metallothionein (Zn7-MT) on the proliferation of NIH3T3 cells induced by hydrogen peroxide inside and outside cells, MTT cell counting method, 3H-TdR incorporation assay, cell cycle phase Determination and other methods. The results showed that menadione and hydrogen peroxide at low concentrations promoted the proliferation of NIH3T3 cells, increased the MTT count, increased the proportion of S phase in cell cycle, and increased the DNA synthesis rate. Zinc 7-metallothionein itself has no effect on NIH3T3 cell proliferation, but it inhibits the effects of menadione and hydrogen peroxide on the proliferation of NIH3T3 cells. It is suggested that zinc 7-metallothionein may play an important role in preventing diseases caused by reactive oxygen species-induced proliferation of fibroblasts.
