目的 通过研究幽门螺旋杆菌对基质金属蛋白酶表达水平的影响 ,探讨幽门螺旋杆菌致病的分子机理。方法 ATCC495 0 3 (CagA+ )、HP0 3 0 92 9(CagA-)幽门螺旋杆菌分别与BGC -82 3共同培养后提取总RNA ,用RT -PCR方法分析BGC -82 3细胞中MMP -9的表达水平 ;通过胃镜活检选取 9例HP阳性胃溃疡、9例HP阴性胃溃疡患者的标本采用RT -PCR分析MMP -9的表达水平 ;取 17例HP阳性和 14例HP阴性的胃溃疡标本经切片后石蜡包埋 ,用免疫组化染色检测MMP -9表达并图象定量分析测量MMP -9染色阳性的面积。结果 HP能诱导MMP -9在BGC -82 3细胞中表达 ,CagA+ HP株明显强于CagA-株 ;在HP阳性胃溃疡的胃黏膜上皮细胞中 ,MMP -9表达水平明显高于HP阴性胃溃疡。结论 HP诱导MMP -9的表达在胃溃疡、胃癌的病理过程中可能起到重要的作用
Objective To study the effect of Helicobacter pylori on the expression of matrix metalloproteinase (MMP) and to explore the molecular mechanism of H. pylori pathogenesis. Methods The total RNA was extracted from ATCC495 0 3 (CagA +) and HP0 3 0 92 9 (CagA-) Helicobacter pylori co-cultured with BGC-82 3, and the expression of MMP-9 in BGC-82 3 cells was analyzed by RT- 9 cases of HP-positive gastric ulcer were selected by gastroscopic biopsy. The expression of MMP-9 in 9 cases of HP-negative gastric ulcer was analyzed by RT-PCR. Seventeen cases of HP-positive and 14 cases of HP-negative gastric ulcer were sectioned, After paraffin embedding, the expression of MMP-9 was detected by immunohistochemical staining and the area of MMP-9 positive staining was quantified by image quantitative analysis. Results HP could induce the expression of MMP-9 in BGC-82 3 cells, and CagA + HP strain was significantly stronger than CagA-strain. The expression of MMP-9 in gastric mucosal epithelium of HP-positive gastric ulcer was significantly higher than that of HP-negative gastric ulcer . Conclusion HP induced MMP-9 expression may play an important role in the pathogenesis of gastric ulcer and gastric cancer
