目的探讨72h睡眠剥夺对雄性小鼠的生殖毒性效应及机制。方法 24只雄性小鼠,随机分为对照组(n=12)、实验组(n=12),经改良多平台睡眠剥夺法睡眠剥夺72h后,镜下观察精子活率、形态及睾丸病理结构,试剂盒测定丙二醛(MDA)、还原型谷胱甘肽(GSH)、超氧化物岐化酶(SOD)值。结果 72h睡眠剥夺后,小鼠精子活率降低,曲细精管上皮萎缩,MDA值、GSH值升高,SOD值降低。结论 72h睡眠剥夺可引起雄性小鼠的生殖毒性效应,这一效应可能与睾丸组织内氧化应激反应增加、抗氧化能力下降并引起睾丸组织的过氧化损伤有关。
Objective To investigate the reproductive toxicity and mechanism of 72h sleep deprivation in male mice. Methods Twenty-four male mice were randomly divided into control group (n = 12) and experimental group (n = 12). After sleep deprivation for 72 hours by modified multi-platform sleep deprivation method, the sperm motility, morphology and histopathology The contents of malondialdehyde (MDA), reduced glutathione (GSH) and superoxide dismutase (SOD) were measured by kit. Results After 72 h sleep deprivation, the sperm motility of mice decreased, the atrophy of seminiferous tubules epithelium atrophy, the MDA value and GSH value increased, and the SOD value decreased. Conclusion 72h sleep deprivation can cause reproductive toxicity in male mice. This effect may be related to the increase of oxidative stress response, the decrease of antioxidant capacity and the damage of testicular tissue peroxidation in testis.
