本文通过研究丁香酚(Eugenol)与黄芩(Scutellaria baicalensis)、葡萄籽(Grape seeds)、五倍子(Chinese gall)、柠檬(Lemon)的提取物对变形链球菌(Streptococcus mutans)及其生物被膜的药效,旨在从中筛选出具有防龋潜能的提取物。采用微量肉汤法测定5种天然药物的最低抑菌浓度(Minimal inhibitory concentration,MIC)、最低杀菌浓度(Minimal bactericidal concentration,MBC)、最低抑制生物被膜浓度(Minimal biofilm inhibitory concentration,MBIC);微量稀释棋盘法测定任两种提取物联合使用时的分级抑菌浓度指数(Fractional inhibitory concentration indexes,FICI);构建生物被膜体外模型,测得提取物对生物被膜生成量的影响;计算各提取物作用生物被膜后被膜内的存活菌数量,并绘制杀菌曲线。丁香酚/柠檬的MBIC/MIC为1~2,在5种提取物中较小;丁香酚、五倍子及柠檬提取物在抑制生物被膜生成方面的效果明显优于黄芩和葡萄籽提取物;杀菌曲线显示柠檬、丁香酚杀菌效果较好,且二者具有一定的协同作用,可以将二者联合使用防龋。
In this paper, we investigated the effects of Eugenol and Scutellaria baicalensis, Grape seeds, Chinese gall and Lemon on Streptococcus mutans and their biofilm , Aimed at screening out the caries potential of the extract. The minimal inhibitory concentration (MIC), minimum bactericidal concentration (MBC) and minimum biofilm inhibitory concentration (MBIC) of five natural medicines were determined by the micro broth method. The microdilution Fractional inhibitory concentration indexes (FICI) were determined by the chessboard method when any two extracts were used in combination. The biofilm in vitro model was established and the effect of the extract on the biofilm production was measured. The number of viable cells in the envelope after coating and the sterilization curve were plotted. The MBIC / MIC of eugenol / lemon was 1 ~ 2, which was smaller among the 5 extracts. The effects of eugenol, gallnut and lemon extract on the inhibition of biofilm formation were significantly better than that of scutellaria baicalensis and grape seed extract. Lemon, eugenol bactericidal effect is better, and the two have a synergistic effect, the two can be combined use of anti-caries.
