目的:探讨磷脂酰肌醇3-激酶/蛋白激酶B(PI3K/Akt)信号转导通路在丹蒌片治疗动脉粥样硬化(AS)中的作用。方法:雄性日本大耳白兔30只随机分为空白对照组、高脂组、可定组、丹蒌片组、丹蒌片+可定组,每组6只。空白对照组饲喂普通饲料,高脂组在普通饲料中加入2%胆固醇及0.02%蛋氨酸;可定组、丹蒌片组、丹蒌片+可定组在普通饲料中加入2%胆固醇及0.02%蛋氨酸及相应的对照药物,复制家兔动脉粥样硬化病变模型。9周后取胸主动脉上段,苏木素-伊红(HE)染色法观察病理组织学改变;Western blot检测PI3K、p-Akt蛋白表达。结果:丹蒌片组、丹蒌片+可定组与高脂组比较AS病变明显延缓;丹蒌片组、丹蒌片+可定组PI3K及p-Akt蛋白表达水平均明显下调,与高脂组相比差异显著(P<0.05,P<0.01)。结论:丹蒌片可通过抑制PI3K/Akt信号转导通路发挥抗AS药理作用。
Objective: To investigate the effect of phosphatidylinositol 3-kinase / protein kinase B (PI3K / Akt) signal transduction pathway in the treatment of atherosclerosis (AS) by danjing tablet. Methods: Thirty male Japanese white rabbits were randomly divided into blank control group, high fat group, fixed group, danji tablet group and danji tablet group (6 rats in each group). Blank control group fed normal feed, high-fat group added 2% cholesterol and 0.02% methionine in ordinary feed; can be set, Dan Dan tablet group, Dan Dan tablet + can be set in ordinary feed by adding 2% cholesterol and 0.02 % Methionine and the corresponding control drug to replicate the model of atherosclerosis in rabbits. After 9 weeks, the upper thoracic aorta was taken and the histopathological changes were observed by hematoxylin-eosin (HE) staining. The protein expressions of PI3K and p-Akt were detected by Western blot. Results: The expression of PI3K and p-Akt protein in Danji tablet group, Danxuan tablet group and Danzhipi tablet group were significantly decreased compared with those in high fat group, There was significant difference between the two groups (P <0.05, P <0.01). Conclusion: Danjian tablet can exert anti-AS pharmacological effects by inhibiting the PI3K / Akt signal transduction pathway.
