目的了解西宁市屠宰场生猪小肠结肠炎耶尔森菌分布及病原学特征。方法从猪咽拭子和回盲部肠内容物标本中提取细菌基因组DNA,进行小肠结肠炎耶尔森菌保守基因foxA的PCR检测,对阳性标本进行细菌分离,阳性菌株进行血清学分型、生物分型和毒力基因检测。结果 400份标本PCR扩增foxA基因阳性72份,其中49份培养分离出小肠结肠炎耶尔森菌,检出率为12.25%(49/400);49株小肠结肠炎耶尔森菌经生化鉴定主要为3个生物型:1A型(占16.33%)、3型(占79.59%)和4型(占4.08%);血清型以O∶3、O∶5、O∶8、O∶9为主,其中O∶3血清型39株(占79.59%),O∶5血清型5株(占10.20%),O∶8血清型2株(占4.08%),O∶9血清型1株(占2.04%),血清不确定型2株(占4.08%)。毒力基因分布特征为ail+、ystA+、ystB-、yadA+、virF+占73.46%(36/49),ail+、ystA+、ystB-、yadA-、virF-占10.20%(5/49),其他毒力基因型占16.33%(8/49)。结论西宁市生猪携带的小肠结肠炎耶尔森菌主要为致病性菌株,应加强对本地生猪小肠结肠炎耶尔森菌的监测。
Objective To understand the distribution and etiological characteristics of Yersinia enterocolitica in live pigs in slaughterhouse in Xining. Methods Bacterial genomic DNA was extracted from swine throat swabs and ileocecal intestinal contents and PCR was performed on foxA gene of Yersinia enterocolitica. The positive samples were isolated by bacteria. The positive strains were serologically genotyped, Typing and virulence gene testing. Results Fusobacterium yersinia was isolated from 49 strains of Yersinia enterocolitica, with a positive rate of 12.25% (49/400). Forty-nine strains of Yersinia enterocolitica were biochemically expressed There were three biotypes identified as type 1A (16.33%), type 3 (79.59%) and type 4 (4.08%). Serotypes were O: 3, O: 5, O: 8, O: 9 Of which, O: 3 serotypes accounted for 79.59%, O: 5 serotypes (10.20%), O: 8 serotypes (4.08%), O: 9 serotypes (2.04%) and 2 (4.58%) of the undefined serotypes. The distribution of virulence genes was ail +, ystA +, ystB-, yadA +, virF + accounted for 73.46% (36/49), ail +, ystA +, ystB-, yadA- and virF- accounted for 10.20% (5/49) Accounting for 16.33% (8/49). Conclusion Yersinia enterocolitica is the main pathogenic strain in live pigs in Xining, and the monitoring of Yersinia enterocolitica in local pigs should be strengthened.
