B_3HM细胞系可以致小鼠白血病。为了进一步寻找致病物质,将B_3HM细胞经超声波匀浆后,差速离心分成四个组分,分别注射于经~(137)Cs 4.5 Gy照射的615或BALB/c小鼠。结果显示,第3组分含致病活性物质。经与B_3HM细胞第3组分共孵育后的MO7e细胞对GM-CSF依赖性明显减弱,并能生成自发性L-CFU。在SDS-PAGE胶上B_3HM细胞第3组分显示一条分子量约36kD的蛋白带。经制备电泳分离的5条蛋白带中3条带对GM-CSF因子依赖细胞系MO7e有一定促增殖作用。B_3HM细胞使经照射处理的小鼠血细胞发生恶性转化的活性物质可能与这些表达蛋白有关。
B_3HM cell line can cause mouse leukemia. In order to further search for pathogenic substances, B 3 HM cells were homogenized by ultrasound and then differentiated into four fractions by differential centrifugation, and were respectively injected into 615 or BALB / c mice irradiated with 4.5 Gy 137 Gy. The results show that the third component contains pathogenic active substances. MO7e cells co-incubated with the third component of B_3HM cells significantly weakened GM-CSF, and could generate spontaneous L-CFU. The third component of B_3HM cells on SDS-PAGE gel shows a protein band of about 36 kD in molecular weight. Three of the five protein bands isolated by preparative electrophoresis had a certain pro-proliferation effect on GM-CSF-dependent cell line MO7e. B_3HM cells may cause malignant transformation of the irradiated mouse blood cells active substances associated with these expressed proteins.
