目的探讨Gαi2在单侧输尿管梗阻(UUO)大鼠肾组织中的表达变化及意义。方法选用36只SD大鼠,随机分为UUO模型组(n=18)以及对照组(sham组,n=18)。分别于术后3、7、14d处死(每个时间点n=6),分成UUO3d组、UUO7d组和UUO14d组。取左肾组织留存。分别采用HE染色和Masson染色法观察每个时间点肾脏组织病理变化,用免疫组织化学法和Western blot法检测每个时间点肾脏组织中Gαi2和Ⅰ型胶原的表达。结果 HE和Masson染色结果显示,随着梗阻时间延长,大鼠肾组织损伤逐渐加重,胶原沉积逐渐增多。免疫组织化学染色结果显示随着梗阻时间延长,大鼠肾脏组织中的Gαi2和Ⅰ型胶原含量逐渐增多,UUO组各组之间以及与对照组相比差异显著(P<0.05)。Western blot结果显示,随着梗阻时间延长,大鼠肾脏组织中的Gαi2和Ⅰ型胶原表达逐渐增高,UUO组各组之间以及与对照组相比差异显著(P<0.05)。相关性分析结果显示,随着大鼠肾脏梗阻时间延长,Gαi2和Ⅰ型胶原表达均与UUO大鼠肾间质胶原沉积呈正相关,Gαi2与Ⅰ型胶原表达呈正相关。结论在单侧输尿管梗阻大鼠肾脏组织中Gαi2和Ⅰ型胶原表达呈时间依赖性,随着梗阻时间延长二者含量逐渐增多,Gαi2可能在肾间质纤维化的发生发展过程中起到重要作用。
Objective To investigate the expression and significance of Gαi2 in renal tissues of unilateral ureteral obstruction (UUO) rats. Methods Thirty-six SD rats were randomly divided into UUO model group (n = 18) and control group (sham group, n = 18). They were sacrificed on days 3, 7 and 14 after operation (n = 6 at each time point) and divided into UUO3d group, UUO7d group and UUO14d group. Left kidney tissue retained. The pathological changes of kidney were observed at each time point by HE staining and Masson staining respectively. The expression of Gαi2 and type Ⅰ collagen in kidney tissues were detected by immunohistochemistry and Western blot. Results The results of HE and Masson staining showed that with the prolongation of obstruction, the injury of rat kidney gradually increased and the deposition of collagen gradually increased. Immunohistochemical staining results showed that with the prolongation of obstruction, the content of Gαi2 and type Ⅰ collagen in rat kidney gradually increased. There was significant difference between UUO group and control group (P <0.05). The results of Western blot showed that the expression of Gαi2 and type Ⅰ collagen in rat kidney increased gradually with the prolongation of obstruction, and there was significant difference between UUO group and control group (P <0.05). Correlation analysis showed that with the prolonged renal obstruction in rats, Gαi2 and type Ⅰ collagen expressions were positively correlated with collagen deposition of renal interstitium in UUO rats, and Gαi2 was positively correlated with type Ⅰ collagen expression. Conclusions The expressions of Gαi2 and type Ⅰ collagen in renal tissues of unilateral ureteral obstruction rats are time-dependent. The content of Gαi2 gradually increases with the prolongation of obstruction, and Gαi2 may play an important role in the development of renal interstitial fibrosis .
