目的探讨莱菔硫烷对人脑胶质瘤细胞(U251)细胞生长的影响及作用机制。方法采用噻唑蓝(MTT)法研究不同浓度的莱菔硫烷对U251细胞生长的抑制作用并测定其半数抑制浓度(IC50);采用Westernblot研究莱菔硫烷对U251细胞内Cyt-c的表达情况。结果在较低剂量范围内(≤100μmol/L),随着作用剂量的增加,莱菔硫烷对U251细胞的生长的抑制作用也明显增强。莱菔硫烷作用72h后的IC50值为(52.41±6.63)μmol/L;莱菔硫烷作用48h后胞浆内Cyt-c的表达量逐渐增多,呈剂量依赖关系。结论莱菔硫烷对U25l细胞具有明显的增生抑制及诱导凋亡作用,其作用机制可能是通过诱发细胞内Cyt-c的释放,引发经线粒体途径的内源性凋亡来实现的。
Objective To investigate the effects of sulforaphane on the growth of human glioma cells (U251) and its mechanism. Methods Thiazolyl tetrazolium (MTT) method was used to study the inhibitory effect of sulforaphane on the growth of U251 cells at different concentrations. The inhibitory concentration (IC50) of U251 cells was determined by MTT assay. The Cyt-c expression of sulforaphane in U251 cells was detected by Western blot. Results In the lower dose range (≤100μmol / L), the inhibitory effect of sulforaphane on the growth of U251 cells was also significantly enhanced with the increase of the dose. The IC50 value of sulforaphane after 72h was (52.41 ± 6.63) μmol / L, and the cytosolic Cyt-c expression gradually increased in a dose-dependent manner after 48 hours of sulforaphane. CONCLUSION Sulforaphane significantly inhibits the proliferation of U251 cells and induces apoptosis. Its mechanism may be through the release of Cyt-c in cells and the initiation of endogenous apoptosis via mitochondrial pathway.
