目的:探讨肾母细胞瘤过表达基因(NOV CCN3)在肝癌中的表达及在肝癌侵袭中的作用。方法 :荧光定量PCR(realtime PCR)检测肝癌及癌旁组织中的CCN3的m RNA表达水平,同时检测肝癌细胞系及正常人肝细胞中CCN3的m RNA表达水平,Western blot检测肝癌及癌旁组织中CCN3蛋白的表达量;通过肝癌细胞过表达CCN3分析CCN3蛋白对肝癌细胞转移的作用;运用Transwell侵袭实验和划痕迁移实验研究CCN3过表达对肝癌细胞侵袭和迁移能力的影响。结果:50对样本中有39对CCN3的m RNA表达水平在肝癌组织中高于癌旁组织,Western blot提示肝癌组织中CCN3蛋白表达量高于癌旁组织。体外肝癌细胞的功能试验中,过表达CCN3能促进MHCC-97H、SMMC-7721细胞的侵袭与转移。结论:CCN3在肝癌组织中高表达,过表达CCN3能促进肝癌的侵袭与转移,CCN3发挥作用机制的研究能为肝细胞肝癌的治疗提供新的思路。
Objective: To investigate the expression of NOV CCN3 in hepatocellular carcinoma (HCC) and its role in the invasion of hepatocellular carcinoma (HCC). Methods: Real-time PCR was used to detect the mRNA expression of CCN3 in hepatocellular carcinoma and its adjacent tissues. Meanwhile, the mRNA expression of CCN3 in hepatocellular carcinoma cells and normal human hepatocytes was detected. Western blot was used to detect the expression of CCN3 in hepatocellular carcinoma and its adjacent tissues CCN3 protein expression in liver cancer cells by CCN3 overexpression of CCN3 protein on the liver cancer cell metastasis; Transwell invasion assay and scratch migration assay CCN3 overexpression on the invasion and migration of hepatocellular carcinoma cells. Results: The m RNA expression of 39 pairs of CCN3 in 50 pairs of samples was higher than that in paracancerous tissues in HCC tissues. Western blot showed that the expression of CCN3 protein in HCC tissues was higher than that in paracancerous tissues. In the function test of hepatoma cells in vitro, overexpression of CCN3 can promote the invasion and metastasis of MHCC-97H and SMMC-7721 cells. CONCLUSION: CCN3 is overexpressed in HCC tissues. Overexpression of CCN3 can promote the invasion and metastasis of HCC. The mechanism of action of CCN3 can provide new ideas for the treatment of hepatocellular carcinoma.
