目的:观察压迫和减压后大鼠背根节(DRG)组织中细胞凋亡及凋亡相关蛋白Bcl-2的表达变化,进一步探讨益气化瘀补肾方对上述指标的影响。方法:选用3月龄SPF级SD雄性大鼠50只,随机分为5组。分别应用TUNEL法检测DRG神经元凋亡程度,并应用免疫组织化学法检测凋亡相关蛋白Bcl-2的表达变化。结果:同假手术组相比,压迫组和减压组DRG神经元细胞凋亡个数显著增高,均具有统计学差异,P<0.01;减压后,凋亡细胞个数减少,与压迫组相比有显著性差异,P<0.05。压迫损伤后Bcl-2在DRG神经元表达升高,但与假手术组相比无显著性差异,P<0.05;在减压组Bcl-2表达进一步升高,与假手术组和压迫组相比,均有显著性差异,P<0.01或P<0.05。益气化瘀补肾方可显著降低DRG神经元凋亡的个数,促进Bcl-2表达,而在行减压手术后其作用更为显著,明显优于减压组,P<0.01。结论:益气化瘀补肾方可显著增加Bcl-2的表达,减少背根神经元的凋亡,其作用在减压手术后,更为显著。
OBJECTIVE: To observe the changes of apoptosis and expression of Bcl-2 in rat dorsal root ganglion (DRG) after compression and decompression, and to explore the influence of Yiqi Huayu Bushen prescription on these indexes. Methods: 50 male SPF SD rats of 3 months old were randomly divided into 5 groups. The apoptosis of DRG neurons was detected by TUNEL method, and the expression of Bcl-2 protein was detected by immunohistochemistry. Results: Compared with the sham operation group, the apoptosis of DRG neurons in the compression group and decompression group were significantly increased (P <0.01, P <0.01). After decompression, the number of apoptotic cells decreased, P <0.05 compared with the significant difference. The expression of Bcl-2 in DRG neurons was increased after compression injury, but there was no significant difference compared with sham operation group (P <0.05). The expression of Bcl-2 in decompression group was further increased. Compared with sham group and compression group Than that of the control group, P <0.01 or P <0.05. Yiqi Huayu Bushen Decoction can significantly reduce the number of apoptotic neurons in DRG and promote the expression of Bcl-2. However, the effect of Yiqi Huayu Bushen Decoction is more significant than that of decompression group (P <0.01). Conclusion: Yiqi Huayu Bushen Recipe can significantly increase the expression of Bcl-2 and decrease the apoptosis of DRG neurons, which is more significant after decompression surgery.
