以紫玉兰花粉为材料,采用离体萌发法研究不同培养基组分对紫玉兰花粉萌发和花粉管生长的影响,确定花粉萌发的最佳培养条件,并用25/10℃、15/10℃、有水和无水4种条件交互处理观察紫玉兰单花的开花进程。研究表明:(1)紫玉兰花粉萌发的最佳培养基是3%蔗糖+0.003%~0.005%硼酸+0.02%氯化钙;(2)花粉萌发率和花粉管长度达到最大值为6 h;(3)15/10℃处理的紫玉兰花瓣明显比25/10℃的大;(4)有水处理的紫玉兰花瓣比无水处理的花瓣干重大;(5)有水处理的表皮细胞排列整齐且比无水处理的表皮细胞大;(6)紫玉兰花药开裂散粉需在干燥环境下进行。本研究对延长花期、增大花瓣面积、提高其观赏价值以及杂交育种具有重要意义。
The effects of different media components on the pollen germination and pollen tube growth of Magnolia biondii were studied by in vitro germination method. The optimal culture conditions for pollen germination were determined. The optimum culture conditions were determined with 25/10 ℃, 15/10 ℃, There are four conditions of water and anhydrous interaction treatment of the flowering process of the single flowering of Magnolia. The results showed that: (1) The optimal medium for the germination of pollen of Magnolia biondii was 3% sucrose + 0.003% ~ 0.005% boric acid + 0.02% calcium chloride; (2) the pollen germination rate and pollen tube length reached the maximum value of 6 h; (3) The purple petals treated with 15/10 ℃ were significantly larger than that of 25/10 ℃; (4) The purple petals treated with water had a greater dry weight than the petals without water treatment; (5) The water-treated epidermal cells Neat and more than the anhydrous treated epidermal cells; (6) purple magnolia anther dehiscence loose powder to be carried out in a dry environment. This study is of great significance to prolong the flowering period, increase the petal area, enhance its ornamental value and crossbreeding.
