目的了解2007-2008年湖南省流感病毒优势流行毒株A(H3N2)亚型的基因变异特征。方法取病原学监测中分离到的29株A(H3N2)亚型流感病毒毒株,对其血凝素基因进行逆转录-聚合酶链反应(RT-PCR),扩增产物纯化后测序;测序结果与WHO全球流感疫苗株序列进行同源比对,绘制种系发生树。结果2007年的分离株与2007-2008年北半球疫苗株A/Wisconsin/67/05比较,变异位点主要为G50E,S138A,K140I,R142G以及N144D。2008年分离株与北半球疫苗株A/Wisconsin/67/05比较,变异位点主要为G50E,S138A,K140I,L157S;而与2008-2009年北半球疫苗株A/Brisbane/10/07比较显示,变异位点主要为K140I,L157S。结论湖南省2007-2008年A(H3N2)亚型流感流行毒株的HA1基因发生了变异,可能是导致其流行的重要原因。
Objective To understand the genetic variation of influenza A (H3N2) subtype in Hunan Province from 2007 to 2008. Methods Thirty-nine strains of influenza A (H3N2) virus isolated from etiological surveillance were collected and their hemagglutinin gene was reverse transcriptase-polymerase chain reaction (RT-PCR). The amplified product was purified and sequenced. Results Homologous alignment with the WHO Global Influenza Vaccine strain sequence and phylogenetic tree was drawn. Results Compared with A / Wisconsin / 67/05 of northern hemisphere vaccine strain 2007-2008, the isolates of 2007 were mainly G50E, S138A, K140I, R142G and N144D. Compared with A / Wisconsin / 67/05 in northern hemisphere vaccine strain in 2008, the variation sites were mainly G50E, S138A, K140I and L157S. Compared with A / Brisbane / 10/07 in northern hemisphere vaccine strain in 2008-2009, Site mainly K140I, L157S. Conclusion The mutation of HA1 gene of influenza A (H3N2) strains in Hunan Province during 2007-2008 may be the important reason leading to its popularity.
