目的:探讨用超声微泡造影剂介导血管生成素-1(Ang-1)基因治疗兔下肢缺血的可行性。方法:将15只新西兰大白兔左侧股动脉及其分支切断结扎后随机分成3组:A组采用局部注射pEGFP/Ang-1质粒与超声微泡造影剂的混合物并用超声照射;B组采用局部注射pEGFP/Ang-1质粒;C组注射生理盐水作为对照组。治疗4周后进行血管造影,了解新生血管情况,免疫组化方法检测Ang-1的蛋白表达。结果:在侧支循环建立、胞浆棕褐色程度、新生血管方面,A组较多,B组其次,C组较少。结论:用超声微泡介导的Ang-1基因转染,可促进缺血骨骼肌的血管新生和侧支循环建立,为下肢缺血性疾病的基因治疗提供了一种新的途径。
Objective: To investigate the feasibility of using ultrasound microbubble contrast medium to mediate angiogenin-1 (Ang-1) gene therapy in rabbit lower limb ischemia. Methods: The left femoral artery and its branches of 15 New Zealand white rabbits were severed and ligated, and were randomly divided into 3 groups: Group A was injected with a mixture of pEGFP / Ang-1 plasmid and ultrasound microbubble contrast agent in local anesthesia; Injection of pEGFP / Ang-1 plasmid; Group C injection of saline as a control group. After 4 weeks of treatment, angiography was performed to understand the neovascularization and immunohistochemistry to detect the protein expression of Ang-1. Results: In the setting of collateral circulation, the degree of cytoplasm brown, neovascularization, A group more, B group secondly, C group less. CONCLUSION: Ang-1 gene transfection mediated by ultrasound microbubbles can promote the angiogenesis and collateral circulation of ischemic skeletal muscle and provide a new way for the gene therapy of ischemic diseases of the lower extremities.
