目的通过比较正常孕妇和重度子痫前期患者胎盘组织vasohibin和VEGF的表达变化及相互关系,探讨vasohibin及VEGF在子痫前期的表达及意义。方法采用免疫印迹法对30例正常妊娠胎盘组织和30例重度子痫前期患者胎盘组织中vasohibin和VEGF蛋白的相对表达量进行检测,运用免疫组织化学法对vasohibin在胎盘组织中进行定位测定。结果正常对照组和重度子痫前期组胎盘vasohibin蛋白的相对表达量分别为0.91±0.12和0.58±0.09,VEGF蛋白的相对表达量分别为0.65±0.20和0.24±0.13,两组比较vasohibin及VEGF的表达差异均具有统计学意义(P<0.05),且二者具有正相关关系(r=0.262,P<0.05);vasohibin选择性表达于胎盘血管内皮细胞,未见合体滋养细胞、细胞滋养细胞和绒毛间质细胞等其他细胞组分有vasohibin阳性表达。结论重度子痫前期患者胎盘组织vasohibin及VEGF的表达降低可能与重度子痫前期的发生有关。
Objective To investigate the expression and significance of vasohibin and VEGF in preeclampsia by comparing the changes of vasohibin and VEGF in normal pregnant women with severe preeclampsia. Methods The relative expression of vasohibin and VEGF protein in placenta from 30 normal pregnant women and 30 severe preeclampsia patients was detected by immunoblotting. The localization of vasohibin in placenta was determined by immunohistochemistry. Results The relative expression levels of vasohibin protein in normal control group and severe preeclampsia group were 0.91 ± 0.12 and 0.58 ± 0.09, respectively, and the relative expression of VEGF protein were 0.65 ± 0.20 and 0.24 ± 0.13 respectively (R = 0.262, P <0.05); vasohibin was selectively expressed in placental vascular endothelial cells, and no syncytiotrophoblast, cytotrophoblast and Vasohibin positive expression of other cell components such as villi interstitial cells. Conclusion The decreased expression of vasohibin and VEGF in severe preeclampsia may be related to the occurrence of severe preeclampsia.
