目的研究人工光环境致生殖节律失调大鼠卵巢的病理机制,以及滋阴补阳中药调整生殖节律的局部作用机理。方法采用持续人工光源环境对SD雌性大鼠持续光照造模,中药组在造模成功后连续用药18日后取卵巢进行HE染色观察卵巢结构,采用免疫组化法、检测隐色素2、褪黑素受体1、黄体生成素受体、表皮生长因子受体。结果持续人工光源环境下生长的雌性大鼠卵巢呈现多囊样改变,各级发育卵泡消失,窦卵泡发育不良,排卵障碍,原位黄素化,形成结构异常、筛网状黄体。滋阴补阳中药上调隐色素2、褪黑素受体1、表皮生长因子受体,并促进了卵泡发育。结论人工光源持续照射可导致卵巢结构和功能异常,滋阴补阳中药可通过上调颗粒细胞及卵泡膜细胞隐色素、褪黑素受体、表皮生长因子受体表达,增强褪黑素在卵巢局部的生物学作用,改善颗粒细胞、卵泡膜细胞激素合成、代谢过程,促进卵泡发育和卵巢结构、黄体功能的改善。
Objective To study the pathological mechanism of ovariectomy induced by reproductive rhythm disorders in artificial light environment and to study the local mechanism of regulating the reproductive rhythm of Ziyin Buyang Chinese medicine. Methods The rats were continuously exposed to artificial light for continuous light modeling. The rats in the Chinese herbal medicine group were treated for 18 consecutive days after the model was established and the ovaries were taken for HE staining to observe the ovarian structure. Immunohistochemistry was used to detect the changes of the content of the latent pigment 2, melatonin Receptor 1, luteinizing hormone receptor, epidermal growth factor receptor. Results The ovaries of female rats growing under artificial light source environment showed polycystic changes, follicles disappeared at all levels, antral follicular dysplasia, ovulation disorders, luteinization in situ, the formation of structural abnormalities, mesh-like luteal. Ziyin Buyang Chinese up-regulation of the pigment 2, melatonin receptor 1, epidermal growth factor receptor, and promote follicular development. Conclusion Continuous exposure of artificial light sources can lead to abnormal ovarian structure and function. Ziyin Buyang Chinese medicine can enhance the expression of melanin, melatonin receptor and epidermal growth factor receptor in granulosa cells and orca cells, Biological effects, improve granulosa cells, follicular cytokine synthesis, metabolic processes, and promote follicular development and ovarian structure, corpus luteum function improved.
