目的 探讨AT_2受体在诱导肾小管上皮细胞凋亡中的作用。方法 通过取新生大鼠肾小管上皮细胞体外培养,传代后加入不同浓度AT_2受体兴奋剂CGP-42112A,在不同时间用流式细胞仪测定凋亡细胞比值,并结合TUNEL分析。结果CGP-42112A不同浓度加入细胞24h后,随着剂量的增大,凋亡细胞所占比值逐渐增加,与对照组比较均具有显著差异,P<0.001。10~(-5)M CGP-42112A作用于细胞,随着时间的延长,凋亡细胞所占比值逐渐增加,与对照组比较有显著差异,P<0.001。结论 AT_2受体可诱导新生大鼠肾小管上皮细胞凋亡,凋亡细胞数与受体剂量正相关,与受体兴奋剂作用时间正相关。
Objective To investigate the role of AT_2 receptor in inducing renal tubular epithelial cell apoptosis. Methods Rat renal tubular epithelial cells were cultured in vitro and subcultured with different concentrations of AT_2 receptor agonist CGP-42112A. The ratio of apoptotic cells was determined by flow cytometry at different time points and analyzed by TUNEL. Results After treated with different concentrations of CGP-42112A for 24h, the percentage of apoptotic cells increased with the increase of dose, which was significantly different from the control group (P <0.001.10 ~ (-5) M CGP-42112A Acting on the cells, with the extension of time, the proportion of apoptotic cells gradually increased, compared with the control group were significantly different, P <0.001. CONCLUSION: AT 2 receptor can induce the apoptosis of renal tubular epithelial cells in neonatal rats. The number of apoptotic cells is positively correlated with the dose of receptor and positively correlated with the action time of receptor agonist.
