目的:探讨清络通痹方精简前后的药效学作用及影响机制。方法:DBA/1小鼠,复制胶原性关节炎(CIA)模型,随机分为空白组,模型组,清络通痹精简方组(8.4 g·kg-1),清络通痹原方组(13 g·kg-1)及雷公藤多苷片组(9 mg·kg-1),观察一般情况及关节肿胀,ig给药4周后,检测血清中白介素-1β(IL-1β),肿瘤坏死因子-α(TNF-α),抗Ⅱ型胶原抗体(抗CⅡ抗体)含量;踝关节HE染色观察病理学改变及病理评分。结果:与正常组比较,模型组小鼠关节肿胀明显,关节炎评分,TNF-α,IL-1β和抗CⅡ抗体水平明显升高(P<0.01);与模型组比较,清络通痹精简方及原方、雷公藤多苷均能明显抑制小鼠足肿胀,降低关节炎评分,降低血清中TNF-α,IL-1β和抗CⅡ抗体水平(P<0.01,P<0.05);病理学检查提示可减轻关节病理损伤,关节总病理评分降低(P<0.01)。结论:清络通痹精简方将不同性味、不同功效药物合理配伍,治疗效果和作用机制与原方相似。
Objective: To explore the pharmacodynamic effect and mechanism of Qingluotongbi before and after streamlining. Methods: DBA / 1 mice were randomly divided into blank control group, model group, Qingluotongbi decoction group (8.4 g · kg -1), Qingluotongbi group (13 g · kg-1) and Tripterygium wilfordii tablet (9 mg · kg-1). The general condition and swollen joint were observed. Serum IL-1β (IL- Tumor necrosis factor-α (TNF-α), anti-type Ⅱ collagen antibody (anti-C Ⅱ antibody) content; ankle joint HE staining pathological changes and pathological score. Results: Compared with the normal group, the joint swelling of the model group was obvious, the arthritis score, the level of TNF-α, IL-1β and anti-CⅡ antibody were significantly increased (P <0.01); Compared with the model group, (P <0.01, P <0.05); Pathology (P <0.01, P <0.05) Inspection tips can reduce joint pathological injury, joint total pathological score decreased (P <0.01). Conclusion: Qingluotongbi concoction will have different taste and reasonable compatibility with different efficacy drugs. The therapeutic effect and mechanism of action are similar to those of the original recipe.
