目的研究黄荆子乙酸乙酯提取物(EVn-50)诱导人乳腺癌MCF-7细胞凋亡作用。方法分3组:空白组(0.02%DMSO)、试验组(EVn-50)、阳性对照组(紫杉醇)。量效测定:试验组给予低、中、高浓度(0.3,3.0,30.0μg·mL-1)的EVn-50,阳性对照组给予紫杉醇(0.2,2.0,20.0μg·mL-1),培养48h。时效测定:2组均给予中浓度,分别培养24,48,72 h。AO/EB双染色荧光显微镜观察凋亡细胞形态;FITC标记Annexin-V/PI双染流式细胞术分析早期凋亡细胞百分率;DNA琼脂糖凝胶蛋白电泳观察MCF-7细胞DNA核小体间断裂的梯形条带。结果在3个时间点Evn-50组均可见细胞凋亡,与紫杉醇组的凋亡率分别为31.83%,47.83%(24 h);51.83%,76.33%(48 h);85.67%,98.5%(72 h)(均P<0.05),低、中、高浓度的Evn-50组与紫杉醇组的凋亡率分别为13%,23.33%(低);51.83%,76.33%(中);55.17%,99.17%(高)(均P<0.05)。DNA在48,72 h有典型梯形条带。结论 EVn-50有诱导人乳腺癌MCF-7细胞凋亡的作用。
Objective To study the apoptosis of breast cancer MCF-7 cells induced by EVn-50. Methods Three groups were divided into blank group (0.02% DMSO), experimental group (EVn-50) and positive control group (paclitaxel). Quantitative measurement: EVn-50 in low, middle and high concentrations (0.3, 3.0, 30.0μg · mL-1) was given to the test group and paclitaxel (0.2,2.0,20.0μg · mL-1) in the positive control group, . Aging: The two groups were given medium concentration, respectively, cultured 24,48,72 h. The morphological changes of apoptotic cells were observed by AO / EB double staining fluorescence microscope. The percentage of early apoptotic cells was analyzed by FITC-labeled Annexin-V / PI double-flow cytometry. The DNA nucleosomes of MCF-7 cells were observed by DNA agarose gel electrophoresis Fractured trapezoidal band. Results The apoptotic rates of Evn-50 group were 31.83%, 47.83% (24 h), 51.83%, 76.33% (48 h), 85.67%, 98.5% (72 h) (all P <0.05). The apoptotic rates of Evn-50 group and paclitaxel group were 13%, 23.33% (low), 51.83%, 76.33% %, 99.17% (high) (all P <0.05). The DNA has a typical trapezoidal band at 48 and 72 h. Conclusion EVn-50 can induce the apoptosis of human breast cancer MCF-7 cells.
