无菌条件下分离提取人胚胎肾小球体外培养,以传代3~5次的系膜细胞,采用3H-胸腺嘧啶核苷( ̄3H-TdR)掺入法和胸腺细胞增殖法观察在不同浓度和不同作用时间雷公藤单体T_4对系膜细胞增殖及其白介素-1(IL-1)产生的影响。结果显示,雷公藤单体T_4明显抑制系膜细胞增殖及IL-1产生,其抑制作用的强弱呈剂量和时间依赖关系。本实验为进一步探讨雷公藤的免疫抑制作用和治疗肾小球疾病的机理提供了新的、有意义的实验依据。
Cultured and isolated human embryonic glomerulus under aseptic conditions in vitro, the mesangial cells were passaged 3 to 5 times. 3H-Thymidine (3H-TdR) incorporation and thymocyte proliferation were used to observe the proliferation of mesangial cells in different concentrations Effects of Tripterygium Wilfordii Monomer T_4 on the Proliferation of Mesangial Cells and the Production of Interleukin-1 (IL-1) at Different Time. The results showed that T_4 of Tripterygium wilfordii inhibited obviously the proliferation of mesangial cells and the production of IL-1, and its inhibitory effect was dose-dependent and time-dependent. This experiment provides new and meaningful experimental evidence for further exploring the immunosuppressive effect of Tripterygium wilfordii and the mechanism of treating glomerular diseases.
