目的 :研究腺苷受体激动剂R -苯异丙基腺苷 (N6-phenyl- 2R -isopropyl-adenosine ,R -PIA)对实验性哮喘豚鼠气道阻力、肺顺应性的影响。方法 :复制豚鼠实验性哮喘动物模型 ,给予R -PIA 5mg/mL的生理盐水雾化吸入 ,观察气道阻力、肺顺应性的变化 ,以及肺泡灌洗液中NO-x 、环一磷酸腺苷 (cAMP)、环一磷酸鸟苷 (cGMP)、肿瘤坏死因子 (TNF)含量的变化。结果 :给予R -PIA雾化吸入后较哮喘组气道阻力增加 (P <0 0 5 ) ,肺顺应性下降 (P <0 0 5 ) ,肺泡灌洗液中NO-x 、cAMP、cGMP、TNF水平下降 ,而TNF含量无明显变化。NO哮喘组为 2 39± 0 0 5 ,R -PIA组为 1 34± 0 0 3(P <0 0 5 ) ,cAMP哮喘组为 1 6 2± 0 0 4,R -PIA组为 0 79± 0 0 2(P <0 0 5 ) ,cGMP哮喘组为 0 47± 0 0 9,R -PIA组为 0 2 5± 0 0 2 (P <0 0 5 ) ,TNF哮喘组为 45 18± 17 2 6 ,R -PIA组为 43 6 2± 14 2 9(P >0 0 5 )。结论 :R -PIA有收缩气道的作用 ,并与肺泡灌洗液中NO-x 、cAMP、cGMP水平下降有关。
Objective: To study the effect of adenosine receptor agonist R-phenyl-2-isopropyl-adenosine (R-PIA) on airway resistance and lung compliance in experimental asthmatic guinea pigs. Methods: The animal model of experimental asthma in guinea pigs was duplicated. The rats were given R-PIA 5mg / mL saline inhalation to observe the changes of airway resistance and lung compliance, and the changes of NO-x and cyclic adenosine monophosphate (cAMP), cyclic guanosine monophosphate (cGMP) and tumor necrosis factor (TNF). Results: Compared with the asthma group, the airway resistance increased (P <0.05), the pulmonary compliance decreased (P <0 05), NO-x, cAMP, cGMP, TNF levels decreased, while no significant changes in TNF content. The NO level in asthma group was 239 ± 05, in R-PIA group was 1 34 ± 0 0 3 (P <0.05), in cAMP asthma group was 1 6 2 ± 0 0 4, in R-PIA group was 0 79 ± 0 0 2 (P 0 05), 0 47 ± 0 0 9 in cGMP asthma group, 0 2 5 ± 0 0 2 in R-PIA group, and 45 18 ± 17 in TNF-asthma group 2 6, and the R-PIA group was 43 6 2 ± 14 2 9 (P> 0.05). Conclusion: R-PIA has the effect of contracting airway and is related to the decrease of NO-x, cAMP and cGMP in bronchoalveolar lavage fluid.
