目的:探讨鳖甲炮制前后抗肝纤维化活性部位的差异,为鳖甲药材的临床用药提供依据。方法:运用SDS-PAGE技术,对生鳖甲与醋鳖甲活性部位进行分析比较;采用MTT比色法测定生鳖甲与醋鳖甲对HSC-T6增殖的影响。结果:炮制后的醋鳖甲所含成分增多,一级谱带相对分子质量降低,导致小分子肽类物质产生;生鳖甲和醋鳖甲均具有抗肝纤维化的作用,但醋鳖甲的药效优于生鳖甲。结论:鳖甲炮制前后活性部位明显差异,为临床选择用药及进一步活性成分研究提供依据。
Objective: To explore the difference of anti-liver fibrosis active fraction before and after turtle shell processing, to provide basis for the clinical use of turtle shell medicine. Methods: SDS-PAGE was used to analyze the active sites of raw turtle shell and vinegar, and the effects of raw turtle shell and vinegar on the proliferation of HSC-T6 were determined by MTT assay. Results: The ingredients of processed vinegar and turtle shell increased, the relative molecular mass of the primary band decreased, resulting in the production of small peptides; raw turtle shell and vinegar and turtle shell have the anti-hepatic fibrosis effect, but vinegar and turtle shell The efficacy is better than raw turtle shell. Conclusion: There are significant differences in the active sites of turtle shell before and after processing, which will provide the basis for clinical choice of drugs and further study of active ingredients.
