目的:建立以反相高效液相色谱(RP-HPLC)法测定鸦胆子油中油酸和亚油酸含量的方法。方法:以ω-溴苯乙酮为衍生化试剂、碳酸钾为碱、N,N-二甲基甲酰胺(DMF)为溶剂进行提取分离,得到衍生化的脂肪酸。色谱柱为Shim-pack VP-ODS(150mm×4.6mm,5μm),流动相为甲醇-乙腈-水(30∶20∶50),检测波长为242nm。结果:油酸和亚油酸的检测浓度分别在0.025~1.0、0.1~0.8mg·mL-1范围内与各自峰面积积分值呈良好线性关系(r分别为0.9996、0.9990);平均回收率分别为98.54%、98.50%,RSD分别为1.81%、1.90%(n均为6)。结论:本方法准确、专属性强,可用于鸦胆子油的质量控制。
Objective: To establish a method for the determination of oleic acid and linoleic acid in Brucea javanica oil by reversed-phase high performance liquid chromatography (RP-HPLC). Methods: The derivatized fatty acids were obtained by using ω-bromoacetophenone as the derivatization reagent, potassium carbonate as the base, and N, N-dimethylformamide as the solvent. The chromatographic column was Shim-pack VP-ODS (150 mm × 4.6 mm, 5 μm). The mobile phase consisted of methanol-acetonitrile-water (30:20:50) and the detection wavelength was 242 nm. Results: The concentrations of oleic acid and linoleic acid showed good linear relationship with the integral values of their peak areas (r = 0.9996, 0.9990, respectively) in the range of 0.025-1.0,0.1-0.8 mg · mL-1. The average recoveries were 98.54%, 98.50%, RSD 1.81%, 1.90% (n is 6). Conclusion: This method is accurate and specific and can be used for the quality control of Brucea javanica oil.
