目的:研究苦参碱、顺铂(DDP)单独及联合应用对人卵巢癌裸鼠移植瘤生长的抑制作用。方法:建立SKOV3细胞裸鼠移植瘤模型,随机分为4组,苦参碱组予苦参碱腹腔注射治疗;DDP组予DDP注射治疗;联合用药组予苦参碱和DDP注射治疗;对照组予生理盐水腹腔注射治疗。连续治疗3周后观察疗效并记录各组裸鼠及其皮下移植瘤生长情况,绘制肿瘤生长曲线,计算肿瘤体积和肿瘤生长抑制率。结果:移植人卵巢癌SKOV3细胞株的裸鼠均能成瘤,成瘤率100%,苦参碱对移植瘤有明显的抑瘤作用,抑瘤率达57.56%。苦参碱组、DDP组、联合用药组治疗后的肿瘤体积及抑瘤率分别与对照组比较,差异均有非常显著性意义(P<0.01),与苦参碱组、DDP组比较,联合用药组肿瘤的体积明显减小(P<0.05),抑瘤率明显增高(P<0.05)。结论:苦参碱对移植瘤有明显的抑瘤作用,苦参碱及顺铂联合应用对移植肿瘤生长抑制作用更强,二者具有协同抗癌作用。
Objective: To study the inhibitory effect of matrine and cisplatin (DDP) alone and in combination on the growth of human ovarian cancer xenografts in nude mice. Methods: The model of SKOV3 cell xenografts in nude mice was established and randomly divided into 4 groups. Matrine group was given intraperitoneal injection of matrine; DDP group was treated with DDP injection; matrine and DDP injection were used in combination group; Intraperitoneal injection of saline for treatment. After 3 weeks of continuous treatment, the curative effect was observed and the growth of nude mice and its subcutaneously transplanted tumors were recorded. The tumor growth curve was drawn and the tumor volume and tumor growth inhibition rate were calculated. Results: Nude mice bearing human ovarian cancer cell line SKOV3 all formed tumors. The rate of tumor formation was 100%. Matrine had obvious antitumor activity on the transplanted tumor and the tumor inhibition rate was 57.56%. Compared with control group, tumor volume and tumor inhibition rate of matrine group, DDP group and combination group were significantly different (P <0.01). Compared with matrine group and DDP group, The volume of tumor in the treatment group was significantly reduced (P <0.05), and the tumor inhibition rate was significantly increased (P <0.05). Conclusion: Matrine has obvious anti-tumor effect on xenograft tumor. Matrine and cisplatin combination have stronger inhibitory effect on the growth of transplanted tumor, both of them have synergistic anticancer effect.
