目的:探讨糖尿病大鼠在中药复方益糖康干预后主动脉NF-κBp65 mRNA含量、蛋白水平表达及NF-κB活化的变化。方法:将50只雄性Wistar大鼠随机分为正常对照组、模型组和中药复方益糖康治疗组。用高脂饲料和链脲佐菌素诱导糖尿病模型,中药煎剂益糖康对中药治疗组进行治疗性灌胃。治疗4周后取大鼠主动脉组织,检测主动脉NF-κBp65 mRNA含量、蛋白水平表达及NF-κB活化情况。结果:中药复方益糖康治疗组能明显降低NF-κBp65 mRNA含量,抑制蛋白水平表达及NF-κB活化,与模型组比较有统计学意义(P<0.01)。结论:益糖康可能通过对炎症核心转录因子NF-κB的影响来降低糖尿病大血管病变的风险。
Objective: To investigate the changes of NF-κB p65 mRNA, protein expression and NF-κB activation in aorta of diabetic rats after intervention of Yikang Kang. Methods: Fifty male Wistar rats were randomly divided into normal control group, model group and Chinese medicine compound Yikang Kang treatment group. Using high-fat diet and streptozotocin induced diabetes model, Chinese medicine decoction Yi Tang Kang Chinese medicine treatment group for intragastric administration. After 4 weeks of treatment, the aorta tissues of rats were taken out and the content of NF-κBp65 mRNA, the expression of protein and the activation of NF-κB in the aorta were detected. Results: The Yikangkang treatment group could significantly decrease the content of NF-κBp65 mRNA, inhibit the expression of protein and the activation of NF-κB, which was statistically significant compared with the model group (P <0.01). Conclusion: Yi Tang Kang may reduce the risk of diabetic macroangiopathy by affecting the nuclear transcription factor NF-κB.
