目的:明确复方水牛角颗粒治疗急性痛风性关节炎的作用机理。方法:一次性大鼠单侧踝关节内注入尿酸钠盐溶液诱导急性痛风性关节炎模型,正常组注射等量生理盐水。造模后4h,分别给予复方水牛角颗粒、秋水仙碱灌胃,正常组和模型组灌胃等量生理盐水。每日1次,共3天,72h取大鼠血清,酶联免疫吸附测定(ELISA)法检测TNF-α、IL-8的蛋白表达。结果:与正常组比较,模型组大鼠血清中TNF-α、IL-8蛋白表达显著升高。与模型组相比,复方水牛角颗粒可明显抑制TNF-α、IL-8的蛋白表达。结论:复方水牛角颗粒具有良好的抗大鼠急性痛风性关节炎作用,其部分作用机制在于:抑制TNF-α、IL-8表达以减少组织的炎性损伤。
Objective: To clarify the mechanism of action of Compound Buffalo Horn Granules in the treatment of acute gouty arthritis. Methods: Acute gouty arthritis model was induced by intraperitoneal injection of sodium urate solution in unilateral ankle joint, and normal saline was injected into normal group. Four hours after model establishment, the compound buffalo horn granules were given respectively, and colchicine was given to the rats in the normal group and the model group by the same amount of normal saline. Serum was collected from rats for 1 day, 3 days and 72 hours respectively. The protein expressions of TNF-α and IL-8 were detected by enzyme linked immunosorbent assay (ELISA). Results: Compared with the normal group, the expression of TNF-α and IL-8 in the model group was significantly increased. Compared with the model group, compound buffalo horn particles can significantly inhibit the protein expression of TNF-α, IL-8. CONCLUSION: Compound Buffalo Hormone Granules has a good anti-gout arthritis effect in rats. Part of the mechanism is that inhibiting the expression of TNF-α and IL-8 can reduce the inflammatory injury of the tissue.
