目的 :比较和评价SDS 聚丙烯酰胺凝胶电泳 (SDS PAGE)结合12 5I示踪法与ELISA法测定血浆中重组人碱性成纤维细胞生长因子 (recombinanthumanbasicfibroblastgrowthfactor,rh bFGF)。方法 :1 0只大鼠iv12 5I rh bFGF1 60 0ng/kg ,用SDS PAGE结合放射性计数测定血浆rh bFGF含量。 9只大鼠ivrh bFGF 1 60 0ng/kg ,用ELISA法测定血浆rh bFGF含量。结果 :两种方法的灵敏度、回收率和重现性好。ELISA法测定血浆rh bFGF含量较SDS PAGE结合12 5I示踪法低。结论 :SDS PAGE结合12 5I示踪法与ELISA均可满足药动学研究的要求
OBJECTIVE: To compare and evaluate the recombinant human basic fibroblast growth factor (rh bFGF) in plasma by SDS polyacrylamide gel electrophoresis (SDS PAGE) combined with 125I tracer and ELISA. Methods: One hundred and twenty rats were injected iv with 60ng / kg iv125I rhBFGF1. The content of rhbFGF in plasma was determined by SDS PAGE combined with radioactive counting. 9 rats ivrh bFGF 1 60 0ng / kg, the content of plasma rh bFGF was measured by ELISA. Results: The sensitivity, recovery and reproducibility of the two methods are good. The level of rhbFGF in plasma was lower than that of SDS PAGE combined with 125I tracer assay. Conclusion: SDS PAGE combined with 125I tracer and ELISA can meet the requirements of pharmacokinetic studies
