目的 通过观察结直肠腺癌中胰岛素样生长因子 -Ⅱ(IGF -Ⅱ)mRNA的表达 ,并结合肿瘤细胞增殖和凋亡状态的研究 ,探讨IGF -Ⅱ基因表达与细胞增殖和凋亡的关系 ,并分析其意义。方法 选取 60例结直肠腺癌石蜡标本 ,采用原位分子杂交法检测IGF -ⅡmRNA ,免疫组化S -P法检测PCNA蛋白 ,TUNEL法检测细胞凋亡 ,多媒体图文分析系统定量肿瘤阳性细胞率。以 15例正常结直肠组织作为对照。结果 结直肠腺癌中IGF -ⅡmRNA阳性细胞率 (3 9 64 %± 7 3 2 % )显著高于正常结直肠组织 (2 3 2 1%± 4 10 % )(P <0 0 1) ;IGF -ⅡmRNA的表达与肿瘤Dukes分期和淋巴结转移有关 ,而与患者的年龄、性别、肿瘤大小、部位和分化程度无关 ;IGF -ⅡmRNA的表达与PCNA、细胞凋亡均呈显著正相关。结论 IGF -Ⅱ可能通过促进细胞增殖和凋亡参与了肿瘤的进展。
Objective To investigate the relationship between IGF-Ⅱ gene expression and cell proliferation and apoptosis by observing the expression of insulin-like growth factor-Ⅱ (IGF-Ⅱ) mRNA in colorectal adenocarcinoma and combining with the study of tumor cell proliferation and apoptosis. And analyze its meaning. Methods Sixty specimens of colorectal adenocarcinoma were collected. IGF-ⅡmRNA was detected by in situ hybridization. The protein expression of PCNA was detected by immunohistochemical S-P method. Cell apoptosis was detected by TUNEL method. The ratio of tumor-positive cells . 15 cases of normal colorectal tissue as a control. Results The positive rate of IGF-ⅡmRNA in colorectal adenocarcinoma was significantly higher than that in normal colorectal tissue (392% ± 4310%) (P <0.01) The expression of IGF-ⅡmRNA was correlated with the Dukes staging and lymph node metastasis, but not with age, sex, tumor size, location and differentiation. The expression of IGF-ⅡmRNA was positively correlated with PCNA and apoptosis. Conclusion IGF-Ⅱ may be involved in tumor progression by promoting cell proliferation and apoptosis.
