目的 :观察脑神经生长素 (cranial nerve growthine,CNG)对兔眼急性高眼压视网膜神经节细胞 (RGC)损伤的保护作用。方法 :健康成年新西兰大耳白兔 30只 ,随机分为 5组 ,每组 6只 ,均一眼为高眼压模型眼 ,另一眼为自身对照眼 ,每组 3只兔术后每日肌肉注射 CNG0 .2 ml,5组兔分别于制造模型眼后 1、 3、 7、 15和 30天处死 ,应用免疫组织化学 SABC法检测 bcl- 2和 TGF-β1,定量分析 CNG的药物疗效。结果 :CNG应用 3天后 ,bcl- 2基因蛋白过表达 ,应用 7天后 ,TGF- β1过表达 ,差异均有显著性 (P<0 .0 5 )。结论 :急性高眼压可能会启动 RGC凋亡的调控因素 ,最终导致 RGC凋亡乃至死亡 ;CNG可在一定程度上抑制 RGC的凋亡。
Objective: To observe the protective effect of cranial nerve growth factor (CNG) on the injury of acute ocular hypertension retinal ganglion cells (RGC) in rabbits. Methods: Thirty healthy adult New Zealand white rabbits were randomly divided into 5 groups with 6 in each group, each with high intraocular pressure (IOP) eyes and the other with self - control eyes. Three rabbits in each group received daily intramuscular injection CNG0.2 ml, 5 rabbits were sacrificed at 1, 3, 7, 15 and 30 days after the model eyes were made. The bcl-2 and TGF-β1 were detected by immunohistochemical SABC method. Results: After 3 days of CNG application, the bcl-2 gene protein was overexpressed. After 7 days of application, TGF-β1 was overexpressed (P <0.05). CONCLUSION: Acute ocular hypertension may activate the regulatory factors of RGC apoptosis, eventually leading to apoptosis and even death of RGC. CNG may inhibit the apoptosis of RGC to a certain extent.
