眠尔通(氨甲丙二酯)一般药典采用酸水解后蒸馏出氨,以酸碱滴定法进行含量测定,方法较为费时且不易获得准确的结果。人们还用分光光度法和气相色谱法测定其含量,但由于经分离提取及纯化后的检材不可避免地要损失一些眠尔通,使测定的含量偏低,而且操作麻烦。1977年米氏(Michotle)等提出酸水解眠尔通后应用氨气敏电极进行含量测定。我们结合办案,试验摸索了硫酸水解眠尔通,应用离子选择电极、格氏作图法快速测定的方法,现结合案例介绍如下:
The general pharmacopoeia of Mendon (Propofol) is distilling out ammonia after acid hydrolysis, and the content is determined by acid-base titration. The method is more time-consuming and difficult to obtain accurate results. People also use spectrophotometry and gas chromatography to determine its content, but due to the separation of the extracted and purified samples inevitably lose some of the messenger, the determination of the content is low, and the operation is cumbersome. In 1977 Michal (Michotle) put forward the acid hydrolysis of Mellon after the application of ammonia gas sensing electrode. We combine case handling, trial groping sulphate hydrolysis of Mellon, the application of ion-selective electrode, the rapid determination of the Germans mapping method, are combined with the case described below:
