目的:比较不同转移潜能的肝癌组织的基因表达谱差异,结合以往研究发现的有关肝癌转移相关分子遗传学异常, 以发现与肝癌转移相关基因。方法:应用基因芯片技术,比较不同转移潜能的肝癌组织标本的基因表达谱,并应用RT- PCR方法对差异表达较明显的基因进行了验证。姑果:不伴转移与伴有转移的肝癌组织之间,有989个基因存在差异表达,其中10个位于染色体8p上。选取定位于8p21.3-p22的类纤维蛋白原1基因(FGL1)(fibrinogen-like 1,同义于 HFREP1,LFIRE1),用RT-PCR的方法进行验证,结果证明FGL1在伴有转移的肝癌组织中表达明显下调。结论:不同转移潜能的肝癌组织中基因表达谱存在明显差别。染色体8p上可能存在抑制肝癌转移的基因。FGL1基因可能是一候选肝癌转移抑制基因,值得进一步研究。
OBJECTIVE: To compare the gene expression profiles of HCC tissues with different metastatic potentials and to identify the molecular abnormalities associated with HCC metastasis, which were found in previous studies, to find out the genes related to HCC metastasis. Methods: Gene expression profiles of HCC tissues with different metastatic potentials were compared using gene chip technology. The genes with more obvious differential expression were verified by RT-PCR. Kakudo: 989 genes were differentially expressed between HCC without metastasis and metastasis, of which 10 were located on chromosome 8p. The Fibrinogen-like 1 gene (FGL1) (synonymous with HFREP1 and LFIRE1) located at 8p21.3-p22 was selected and verified by RT-PCR. The results showed that FGL1 was expressed in hepatocellular carcinoma with metastasis Tissue expression was significantly reduced. Conclusion: The gene expression profiles of HCC tissues with different metastatic potentials are significantly different. Chromosome 8p may exist on liver cancer metastasis genes. FGL1 gene may be a candidate gene transfer inhibitor of liver cancer, it is worth further study.
