目的运用磷脂酰肌醇3-激酶(phosphatidylinositol 3-kinase,PI3-K)特异性抑制剂LY294002[2-(4-吗啉基)-8-苯基-4氢-1-苯并吡喃-4-酮]作用于宫颈癌细胞株SiHa,探讨其对宫颈癌细胞增殖的抑制作用。方法培养宫颈癌细胞株SiHa,分为对照组和LY294002干预组(LY294002 5μmol/L、10μmol/L、25μmol/L、50μmol/L)分别作用6h、12h、24h、48h;MTT比色法检测体外使用抑制剂对宫颈癌细胞株SiHa生长的作用;运用流式细胞技术检测SiHa凋亡抑制率。结果 LY294002能抑制宫颈癌体外培养细胞系SiHa的生长,并以剂量-时间依赖方式诱导凋亡,差异有统计学意义(P<0.05)。结论 LY294002能有效抑制SiHa细胞的增殖,呈时间-剂量依赖性。PI3K/Akt介导的信号转导通路可能宫颈癌的发生发展起作用。
OBJECTIVE: To investigate the effect of phosphatidylinositol 3-kinase (PI3-K) inhibitor LY294002 [2- (4-morpholinyl) -8- 4-keto] on cervical cancer cell line SiHa, to explore its inhibition of cervical cancer cell proliferation. Methods The cervical cancer cell line SiHa was cultured and divided into control group and LY294002 intervention group (LY294002 5μmol / L, 10μmol / L, 25μmol / L and 50μmol / L) for 6h, 12h, 24h and 48h respectively. MTT colorimetric assay The effect of inhibitor on the growth of cervical cancer cell line SiHa was tested. The apoptosis inhibition rate of SiHa was detected by flow cytometry. Results LY294002 could inhibit the growth of cervical cancer cell line SiHa in vitro and induce apoptosis in a time-and dose-dependent manner. The difference was statistically significant (P <0.05). Conclusion LY294002 can effectively inhibit the proliferation of SiHa cells in a time-and dose-dependent manner. PI3K / Akt-mediated signal transduction pathway may play a role in the development of cervical cancer.
