目的:探讨聚酰胺对白背三七总黄酮的纯化条件及效果。方法:采用紫外分光光度法测定总黄酮的含量,以总黄酮的保留率和转移率为指标考察上样液pH、上柱吸附流速、树脂药材比、树脂径高比、清洗液pH及流速、洗脱液种类及流速等影响因素。结果:筛选的纯化条件为上样液和清洗液的pH为3.0,树脂药材比2:1,树脂径高比1:8,上柱吸附流速为2BV·h~(-1),70%乙醇为洗脱液,清洗流速为5 BV·h~(-1),洗脱流速为4 BV·h~(-1)。在该纯化条件下,总黄酮含量由粗提物的2.43%提高到24.6%。结论:该纯化条件将白背三七总黄酮的含量提高10倍以上,可为进一步研究提供参考。
Objective: To investigate the purification condition and effect of polyamide on total flavonoids from Panax notoginseng. Methods: The content of total flavonoids was determined by ultraviolet spectrophotometry. The retention rate and transfer rate of total flavonoids were used as indices to investigate the effects of pH value, adsorption rate on column, ratio of resin to drug, resin diameter to height ratio, pH and flow rate, Eluate types and flow rate and other influencing factors. Results: The purification conditions of screening were as follows: the pH of the sample solution and washing solution was 3.0; the ratio of resin to drug was 2: 1; the diameter of resin was 1: 8; the flow rate of adsorption on the column was 2BV · h ~ As eluent, the washing flow rate was 5 BV · h -1 and the elution flow rate was 4 BV · h -1. Under this purification condition, the total flavonoid content increased from 2.43% of the crude extract to 24.6%. Conclusion: This purification condition increased the content of total flavonoids in Panax notoginseng by more than 10 times, which may provide reference for further research.
