目的:建立人血浆中苯溴马隆浓度的HPLC测定方法。方法:色谱柱为Hypersil BDS C_(18)(250 mm×4.6 mm,5μm);流动相为甲醇.水.三乙胺(48:52:0.02),流速1ml·min~(-1);紫外检测波长357 nm;以香豆素为内标,采用乙腈直接沉淀蛋白后进样。结果:苯溴马隆的线性范围为0.01~0.5 mg·L~(-1)(r=0.999 2);提取回收率为103.5%,106.8%,方法回收率为109.5~113.2%,日内和日间RSD分别小于7.0%和9.5%结论:本法操作简便、灵敏、准确,可用于临床治疗度高。
Objective: To establish a method for the determination of benzbromarone in human plasma by HPLC. Methods: The chromatographic column was Hypersil BDS C 18 (250 mm × 4.6 mm, 5 μm). The mobile phase consisted of methanol, water, triethylamine (48:52:.02), flow rate 1 ml · min -1 The detection wavelength was 357 nm. Coumarin was used as internal standard, and the sample was directly precipitated with acetonitrile. Results: The linear range of benzbromarone was 0.01-0.5 mg · L -1 (r = 0.999 2). The recoveries were 103.5% and 106.8%, respectively. The recoveries were between 109.5 and 113.2% Inter-RSD less than 7.0% and 9.5% Conclusion: This method is simple, sensitive, accurate, and can be used for clinical treatment.
