目的 :利用ECHO克隆系统建立一种更加快速、方便、通用的DNA重组技术。方法 :用常规重组法构建出ECHO系统中的携带外源基因HGH的供载体 ,再利用ECHO系统快速构建人生长激素的酿酒酵母表达载体 ,使HGH能在酿酒酵母启动子GAL的调控下进行转录。结果 :基因测序表明 ,在所构建的融合质粒载体上确实含有HGH的基因序列。结论 :本方法为研究HGH的消化道基因治疗打下基础。
Objective: To establish a more rapid, convenient and universal DNA recombination technology using ECHO cloning system. Methods: The vector containing the exogenous gene HGH in ECHO system was constructed by conventional recombinant method, and then the human growth hormone-producing Saccharomyces cerevisiae expression vector was rapidly constructed by using ECHO system so that HGH could be transcribed under the regulation of Saccharomyces cerevisiae promoter GAL . Results: Gene sequencing showed that the constructed fusion plasmid vector did contain HGH gene sequence. Conclusion: This method lays a foundation for studying the gene therapy of gastrointestinal tract in HGH.
