目的:同时测定人工牛黄中胆酸及猪去氧胆酸的含量。方法:以甲醇为提取溶剂,制备供试品溶液;应用薄层色谱法,使用铝基硅胶G高效薄层板,以环己烷-乙醚-冰醋酸(10:5:5)为展开剂,10%磷钼酸乙醇溶液为显色剂;采用单波长吸收扫描法,λ=650 nm,光束狭缝10.0 mm×0.2 mm。结果:胆酸,猪去氧胆酸的线性范围均在0.2~0.6μg之间(r≥0.99);平均回收率(n=6)分别为101.3%(RSD=2.6%)和101.6%(RSD=2.5%)。结论:该方法专属性强,重复性好,适用于人工牛黄中胆酸及猪去氧胆酸的含量测定与质量控制。
Objective: Simultaneous determination of bile acid and hyodeoxycholic acid in artificial bezoar. METHODS: Methanol was used as the extraction solvent to prepare the test solution. Thin layer chromatography (TLC) was performed on a silica gel G high efficiency thin layer plate using cyclohexane-ether-glacial acetic acid (10: 5: 5) 10% phosphomolybdic acid ethanol solution as the color reagent; using single wavelength absorption scanning method, λ = 650 nm, the beam slit 10.0 mm × 0.2 mm. Results: The linear ranges of cholic acid and hyodeoxycholic acid were both 0.2 ~ 0.6μg (r ≥ 0.99). The average recoveries were 101.3% (RSD = 2.6%) and 101.6% (RSD = 2.5%). Conclusion: This method is specific and reproducible. It is suitable for the determination and quality control of bile acid and hyodeoxycholic acid in artificial bezoar.
