目的研究趋化因子CCL19在结直肠肿瘤中的作用。方法采用Real-Time PCR和Western blotting技术筛选高表达CCL19的细胞株SW620细胞作为研究对象,并用siRNA沉默SW620中的CCL19基因。分别采用细胞增殖实验、Transwell迁移、侵袭实验和小管形成实验来检测SW620细胞的增殖、迁移、侵袭和促血管形成能力的变化。结果 Western blotting和Real-Time PCR证实SW620中CCL19相对其他细胞株高表达。经siRNA-CCL19干扰后,SW620细胞在48~120 h内细胞增殖能力显著上升(P<0.05),细胞迁移、侵袭能力亦明显上升(P<0.05),促血管形成能力明显增强。结论 SW620是相对高表达CCL19的细胞株,沉默CCL19基因后可以明显促进SW620细胞在体外的增殖、迁移、侵袭和促血管形成能力,因此认为CCL19在结直肠癌发展中起着抑制作用,并有应用于抗癌药物研究的前景。
Objective To study the role of chemokine CCL19 in colorectal cancer. Methods Real-time PCR and Western blotting were used to screen SW620 cell line which is highly expressed in CCL19 cells and siRNA was used to silence the CCL19 gene in SW620 cells. The proliferation, migration, invasion and angiogenesis of SW620 cells were detected by cell proliferation assay, Transwell migration assay, invasion assay and tubular formation assay. Results Western blotting and Real-Time PCR confirmed that CCL19 was highly expressed in SW620 cells compared with other cell lines. After siRNA-CCL19 interference, the cell proliferation ability of SW620 cells increased significantly (P <0.05) within 48-120 h, and the migration and invasion ability of SW620 cells increased significantly (P <0.05). Conclusion SW620 is a relatively high expression of CCL19 cell line, silencing CCL19 gene can significantly promote SW620 cells in vitro proliferation, migration, invasion and angiogenesis, CCL19 that in the development of colorectal cancer play an inhibitory role, and Applied to anticancer drug research prospects.
